What is PT-141?
PT-141 — also known as Bremelanotide — is a synthetic cyclic peptide derived from Melanotan II. It was discovered when researchers studying MT-II for tanning noticed significant effects on sexual arousal. PT-141 was subsequently developed as a targeted compound for sexual function research, and it went on to achieve full FDA approval under the brand name Vyleesi in 2019.
What makes PT-141 fundamentally different from compounds like Tadalafil (PDE5 inhibitors) is its mechanism — it works through the brain rather than the blood vessels. By activating MC3R and MC4R melanocortin receptors in the hypothalamus, it influences sexual desire and arousal through central nervous system pathways. This means it addresses the desire component of sexual function, not just the physical response.
What Researchers Are Exploring
Sexual Desire & Arousal PT-141’s primary and most extensively studied area is sexual function. Clinical trials demonstrated significant improvements in sexual desire and arousal — in both men and women — through melanocortin receptor activation in the brain. Its FDA approval was based on this research.
Central Nervous System Mechanism Unlike vascular-based compounds, PT-141 works through the brain’s melanocortin system. This central mechanism means it targets the psychological component of sexual function — desire and motivation — rather than just the physical response.
Research in Both Men and Women PT-141 is one of very few compounds studied for sexual function in both sexes. While most sexual dysfunction compounds target male physiology, PT-141’s brain-based mechanism has shown relevant effects in female sexual function research as well — which is part of why it achieved FDA approval for premenopausal women.
Worth Noting PT-141’s FDA approval (as Vyleesi) gives it a clinical credibility that most peptides simply don’t have. It’s the only melanocortin-based peptide to achieve regulatory approval for sexual function, and its unique central mechanism makes it fundamentally different from vascular-based approaches
